When you boot up the console for the first time, you are asked if you want to have voice commands turned on or not. You can change this setting at anytime after the initial set up. If you have voice commands enabled, then it is always waiting for you to say "Xbox" to prompt a voice command, and if voice commands are disabled, then it is not listening for the prompt.

When using voice commands you must preface it by saying, "Xbox" and then the command you want to use. A small microphone appears in the top right corner and it will display the command you issued then perform it. The Xbox Kinect will be in listen mode from then on (I believe it stops listening after a certain short period, I may be wrong.) But in order to have it stop listening when you want, just say so. The wake up feature is not affected by the stop listening command, nor is any other voice activated command. You are simply just telling the Xbox you are done with voice commands at the moment. Next time you say "Xbox" it will begin listening for a command.
